SS-31

What is SS-31?

SS-31 (also known as Elamipretide) is a fascinating "mitochondrial medicine." Unlike most peptides that target hormones or brain chemicals, SS-31 goes straight to the inner membrane of your mitochondria. It’s primarily used in research to repair damaged mitochondria, reduce oxidative stress, and improve ATP (energy) production.

Because SS-31 is often used for "rejuvenation" of the cellular powerplants, its protocol is usually shorter and more intense than a growth hormone peptide.

1. Reconstitution & Dosage
SS-31 is typically found in 50mg vials. Because the effective dose is quite high compared to other peptides, you’ll need more bacteriostatic water for reconstitution.

• Reconstitution: Add 2ml of Bacteriostatic Water to a 50mg vial.
• The Concentration: This creates 25mg per 1ml.
• Syringe Measurement: On a standard U-100 insulin syringe:
o 4 units (0.04ml) = 1mg
o 40 units (0.40ml) = 10mg

Common Research Dosages:

• Standard Dose: 10mg per day.
• Intensive "Repair" Dose: 20mg – 40mg per day (often split into two doses).
• Note: Many researchers start with a "Loading Phase" of 10mg/day to see how the body responds to the sudden increase in mitochondrial efficiency.

2. The Cycle (Duration & Timing)
SS-31 is rarely used as a year-round peptide. It is most effective when used as a "cellular reset."

Goal Typical Cycle Frequency
Mitochondrial Reset 20–30 Days ON Once Daily
Recovery / Post-Illness 10–14 Days ON Once Daily
Maintenance 10 Days ON / 20 Days OFF Once Daily

Pro Tip: Most researchers suggest doing one 20-30 day "Blast" cycle once or twice a year, rather than low-dosing it indefinitely.

3. Timing & Stacking
• Timing: Morning is generally preferred, as the increase in ATP production can be energizing.
• The "Mito-Stack": For maximum results, SS-31 is often followed immediately by a cycle of MOTS-c.
o Think of it like this: SS-31 repairs the factory (mitochondria), and MOTS-c cranks up the production line.

4. Storage & Stability
• Fridge Required: Once mixed, keep it in the refrigerator.
• Stability: SS-31 is relatively stable compared to VIP or Tesamorelin, but it should still be used within 30 days of reconstitution for maximum efficacy.

5. Potential Side Effects
SS-31 is generally considered very safe in clinical trials, but some users report:
• Initial Fatigue: Paradoxically, some feel tired for the first 2-3 days as the body "cleans out" oxidative waste.
• Injection Site Irritation: Because the dose is higher (40 units), ensure you are rotating sites properly using the map we created.
• Increased Endurance: You may notice a lower heart rate during cardio as your cells become more efficient at using oxygen.

Quick Summary for your 50mg Vial: > To hit a 10mg daily dose, you would pull to the 40 unit mark on your syringe (assuming 2ml of water in the vial). One 50mg vial will last you exactly 5 days at this dose.

This "Mitochondrial Restoration" protocol is a popular two-phase approach. Phase 1 (SS-31) repairs the mitochondrial architecture, and Phase 2 (MOTS-c) enhances metabolic flexibility and energy output.
Because SS-31 is usually a daily injection and MOTS-c is typically injected only 1–3 times per week, the calendar looks a bit different for each month.

Phase 1: The "Repair" Month (SS-31)
• Goal: Clean up oxidative stress and fix mitochondrial membranes.
• Schedule: Every Day for 20–25 days.

Sun Mon Tue Wed Thu Fri Sat
SS-31 SS-31 SS-31 SS-31 SS-31 SS-31 SS-31
SS-31 SS-31 SS-31 SS-31 SS-31 SS-31 SS-31
SS-31 SS-31 SS-31 SS-31 SS-31 SS-31 SS-31
________________________________________

Phase 2: The "Activation" Month (MOTS-c)
• Goal: Increase ATP production and glucose metabolism.
• Schedule: 5mg to 10mg, injected 2–3 times per week for 4 weeks.

Sun Mon Tue Wed Thu Fri Sat
Rest MOTS-c Rest MOTS-c Rest MOTS-c Rest
Rest MOTS-c Rest MOTS-c Rest MOTS-c Rest
Rest MOTS-c Rest MOTS-c Rest MOTS-c Rest

Protocol Notes for the Transition:

1. The "Bridge" Week: You don't need a break between the two. In fact, many researchers start MOTS-c the very day after the last dose of SS-31.

2. MOTS-c Reconstitution: MOTS-c is extremely fragile and prone to "gelling."

o Tip: Use at least 2ml of Bacteriostatic Water and let it sit for 5–10 minutes to dissolve on its own. Never shake MOTS-c.

3. Exercise Connection: While SS-31 works while you rest, MOTS-c is a "metabolic shield" that works best if you exercise within 2–4 hours of the injection.

4. Storage: Both of these are highly temperature-sensitive. If you are traveling during this transition, you will need a chilled peptide travel case.

Quick Checklist for your Website/Log:
• [ ] Vials Ready: 4–5 vials of SS-31 (50mg) and 4–8 vials of MOTS-c (5mg or 10mg).
• [ ] Syringes: Since you are injecting daily for Phase 1, make sure you have a full box of 100 syringes.
• [ ] Empty Stomach: Both are best administered in a fasted state (usually first thing in the morning).
