NAD+
What is NAD+?

NAD+ (Nicotinamide Adenine Dinucleotide) isn't technically a peptide—it’s a coenzyme found in all living cells—but in the world of subcutaneous injections, it’s treated much like one. It is the "fuel" for your mitochondria and a key player in DNA repair (via SIRT1 enzymes).
Unlike most peptides, NAD+ injections can be very "intense" if pushed too quickly or at too high a dose. Here is the standard research protocol.

1. Reconstitution & Dosage
NAD+ usually comes in large vials, often 500mg or 1000mg (1 gram), because the effective dose is much higher than that of a traditional peptide.
• Reconstitution: For a 1000mg vial, add 5ml of Bacteriostatic Water.
• The Concentration: This creates 200mg per 1ml.
• Syringe Measurement: On a standard U-100 insulin syringe:
o 5 units (0.05ml) = 10mg
o 25 units (0.25ml) = 50mg
o 50 units (0.50ml) = 100mg
Common Research Dosages:
• Starting Dose: 25mg – 50mg (to assess tolerance).
• Standard Anti-Aging Dose: 100mg – 150mg per day.
• Intensive Recovery Dose: 200mg+ (often administered via IV in clinics, but some do it SubQ).

2. The Cycle (Duration & Timing)
NAD+ levels drop as we age, so many researchers use a "Loading Phase" followed by a "Maintenance Phase."
Phase Duration Frequency
Loading Phase 10–14 Days Once Daily
Maintenance Ongoing 2–3 times per week
The "Blast" 5 Days High dose (150mg+) for rapid recovery
Pro Tip: Always inject in the morning. NAD+ is highly energizing and boosts cellular ATP. If you take it at night, you will likely be staring at the ceiling until 3:00 AM.

3. The "NAD+ Squeeze" (Side Effects)
This is the most important part of the protocol. When you inject NAD+ subcutaneously, you may feel a unique set of temporary side effects known as the "squeeze":
• Chest Tightness: A heavy feeling in the chest (not a heart attack, but it feels like a dull pressure).
• Nausea: A sudden "knot" in the stomach.
• Sinus Pressure: A weird sensation in the bridge of the nose or forehead.
How to avoid it: Inject S-L-O-W-L-Y. Take at least 30–60 seconds to depress the plunger. If you feel the squeeze, stop, wait 20 seconds, and continue.

4. Storage & Stability
• Refrigeration: NAD+ is extremely sensitive to heat. Once reconstituted, it must stay in the fridge.
• Color Check: The liquid should be clear. If it turns noticeably yellow or cloudy, it has likely oxidized and lost potency.

5. The "Synergy" Stack
NAD+ works best when paired with certain "activators":
1. Trimethylglycine (TMG): NAD+ utilizes methyl groups. Taking a TMG supplement (500mg–1000mg) orally helps prevent "methyl depletion," which can cause fatigue in some users.
2. SS-31: As we discussed, SS-31 repairs the mitochondrial "factory," and NAD+ provides the "fuel." They are a match made in cellular heaven.

Quick Summary for your 1000mg Vial:
To get a 100mg dose, you would pull to the 50 unit mark on your syringe (assuming 5ml of water was used to mix). One 1000mg vial will give you 10 doses at this level.
